MOISTURE DETERMINATION
Every crude drug contains some moisture in it, so it is very important to determine the percent of moisture inside it. there are several methods to determine it but the loss on drying is an easy and effective technique. Moisture content determination is one of the important parameters for quality evaluation of crude drug and crude extract. The presence of excess moisture favors the growth of the microorganism and deteriorates the crude drug. So moisture content should be within a specified limit which is acceptable.
To perform loss on drying experiments we have to weigh the crucible in an empty state and then measure 0.50 gm of the crude drug than allow it to dry in 100105 degree for about 15 mint. than place it in a desiccator to cool down and to prevent further moisture absorption and then again place it in the oven till constant weight is received. now finally calculate the moisture content by using the given formula in the video.
Procedure: (Drying in Oven):
Loss on drying is loss of weight expressed as % w/w resulting from the water and volatile matter of any kind that can be driven off under specified condition.This test is carried out on a wellmixed sample of the substance. If the substance is in the form of large crystals, reduce the size by rapid crushing to a powder.
1. Weigh a glass stoppered, shallow weighing bottle (W1) that has been dried under the same conditions to be employed in determination.
2. Transfer to the bottle the quantity of the sample specified in the individual monograph. Cover it and accurately weigh the bottle and the contents (W2).
3. Distribute the sample as evenly as practicable by gentle sidewise shaking to a depth not exceeding 10 mm.
4. Place the loaded bottle in drying chamber as directed in the monograph per and leave it also in the chamber. Dry the sample to constant weigh or for the specified time and at the temperature indicated in the monograph.
5. After drying is completed, open the drying chamber, close the bottle promptly and allow it to cool to room temperature (where applicable) in a desiccator before weighing. Weighing bottle and contents(W3).
6. When “dry to constant weight” is specified in a monograph, drying shall be continued until two consecutive weighings (W4) do not differ by more than 0.50 mg per g of substance taken, the second weighing following an additional hour of drying. Upon opening the chamber, close the bottle promptly, and allow it to come to room temperature in a desiccator before weighing.

Factors affecting moisture content in solid substances:
• Nature of the substance
• Degree of its fragmentation
• Solution in which the substance is formed
• Ambient humidity

Impact of moisture content:
a) Powder flow: Moisture decreases flowabilityof powder by forming stronger interparticle liquid bridges.
 
b) Hardness of tablet: In direct compression process, high and extra low moisture content affects the hardness of tablet.
c) Viscosity: Moisture content changes viscosity of semisolid dosage forms.
d) Growth of microorganisms: Presence of moisture influences growth of microorganisms in solid and semisolid dosage forms.
e) Stability: Presence of moisture influences physical and chemical stability of pharmaceutical dosage forms.
Methods for determination of moisture content:
Karl Fischer moisture meter method
Oven drying method / loss on drying moisture meter method (LOD)
Electrical moisture meter method
Microwave moisture meter method
Nuclear moisture meter method

Oven drying method / loss on drying moisture meter method (LOD):
The original primary moisture measurement method was Loss OnDrying (LOD). In an LOD test, the sample is weighed, dried, and weighed again. The difference in the two weights (Loss on Drying) is then compared with either the original weight (Wetbase test) or final weight (Drybase test) and the moisture content calculated. Tests can be manually conducted (weigh, oven dry, weigh) or automated (integrated weight and heating unit) with systems called Moisture Determination Balances.
Advantages:
It is a primary method (does not require calibration).
Can measure high levels of moisture.
Moisture balances are cheaper than more sophisticated instruments.
Deficiencies:
All volatiles are evaporated, causing improper moisture content calculations.
Operator error can contribute to inaccurate readings.
Air ovens are large and occupy a significant amount of space.
Cannot easily test liquids.
Samples are destroyed or altered.
Tests take some time to conduct.
